Alkyne-Modified Protein Capture using Streptavidin-Free Enrichment involves the selective isolation of proteins labeled with alkyne groups without relying on biotin–streptavidin interactions. Instead, these proteins undergo copper-catalyzed or strain-promoted azide–alkyne cycloaddition (CuAAC or SPAAC) with azide-functionalized affinity tags or solid supports, enabling direct and specific capture. This approach avoids potential issues with endogenous biotin or streptavidin background, offering greater experimental flexibility and cleaner enrichment in proteomics and labeling workflows.
