Biotin-dPEG®₁₂-DBCO
Biotin-dPEG®12-DBCO, product number QBD-11811, is a click chemistry biotinylation reagent built on a medium-length (40 atoms, 46.3 Å) single molecular weight PEG linker with a discrete chain length (dPEG®) and functionalized with dibenzylcyclooctyne (DBCO) for strain-promoted azide-alkyne cycloaddition (SPAAC), also known as “copper-free click chemistry.” The DBCO functional group reacts with any available azide moiety on a target molecule via SPAAC. The amphiphilic dPEG® spacer imparts water solubility to the biotin label, reducing non-specific hydrophobic interactions and reducing or eliminating concerns over aggregation and precipitation due to biotin’s well-known hydrophobicity. Because the hydrophilic dPEG® linker is longer than the classic, hydrophobic LC-LC-biotin, the biotin moiety has potentially better access to the binding pockets of avidin and streptavidin. Peptide and protein modification can be used to introduce suitable azide partners for this biotin labeling compound.
