Recommended Usage
VectaMount Mounting Medium is a non-flammable, non-hazardous, and odorless formula for permanently preserving precipitable enzyme substrates in tissue sections or cell preparations. VectaMount Mounting Medium is compatible with most* clearing agents, including aliphatic hydrocarbon- (e.g. d-limonene) or aromatic hydrocarbon- (e.g. toluene or xylene) based clearing agents.
*For most xylene-free clearing agents such as Clear-Rite 3, incubation times of at least 3x 3-5 minutes is recommended.
In some IHC detection reagent instructions, the last step states “dehydrate, clear and mount”. What does this mean?
In a manual IHC application, these steps involve submersion of the slides into a graded ethanol series, immediately followed by submersion into a miscible solution (xylene or alternative), followed by placement of a coverslip over the preparation with a suitable mounting medium. These steps preserve the specimen for subsequent visualization, imaging and archiving. From our experience, we suggest moving the slides in a slide rack through a series of dishes, commencing dehydration at 95% ethanol (2 changes for 2 min ea.), then into 100% ethanol (2x 2 min ea.), then into clearing solution (3x 2 min ea.), followed by mounting (coverslipping).
VectaMount Mounting Medium has a viscosity which provides for easy application and uniform spreading over the tissue section. Mounted sections are clear with an ideal refractive index suitable for high resolution oil immersion microscopy. VectaMount™ Mounting Medium preserves the color and intensity of preparations stained with enzyme substrates such as DAB, TMB, and BCIP/NBT, as well as our proprietary substrates Vector® NovaRED™ , Vector VIP, Vector SG, ImmPACT® DAB, ImmPACT DAB EqV, ImmPACT NovaRED, ImmPACT VIP, ImmPACT SG, Vector Red, ImmPACT Vector Red, Vector Blue, and Vector Black. The crystal formation that frequently occurs with the alkaline phosphatase substrate BCIP/NBT using other permanent mounting media is essentially eliminated.
