This product is a blocking reagent for in situ hybridization procedures. 1x of this blocking solution is the recommended diluent for fluorescent and enzymatic detection systems used in ISH and FISH. The 100 ml concentrate supplies sufficient reagent to prepare 500 ml of working solution.

Recommended Usage This product is designed to be used as a general blocking agent/diluent for enzyme-based and fluorescent in situ hybridization.Before use, shake well and dilute 5x ISH Blocking Solution to 1x with deionized or distilled water. After hybridization of labeled DNA/RNA probes, tissue sections or chromosome spreads should be blocked for ≥ 30 minutes in 1 x In Situ Hybridization Blocking Solution before application of detection reagents. Detection reagents can be diluted in 1 x blocking solution for approximately 30 minutes before use to further reduce any non-specific binding. Slides can be washed for 2 x 3 minutes in 1x blocking solution between each detection step. For enzyme-based detection, wash tissue sections or chromosome spreads 2 x 5 minutes with TBS* or PBS* before applying substrate. Enzyme substrate should be made according to kit instructions. For fluorescence detection, wash tissue sections or chromosome spreads for 2 x 5 minutes in 4x SSC* + 0.1% Tween® 20 before coverslipping. This solution should not be used with antibodies that cross react with bovine proteins (e.g. anti-goat Ig, anti-sheep Ig).
