Greater sensitivity can be achieved by using an amplified
detection procedure. Our biotin/avidin and biotin/streptavidin
systems introduce a large number of enzymes to the target
site providing signal amplification while maintaining low
background.
The VECTASTAIN® ABC-AmP™ is an amplified ABC alkaline
phosphatase reagent for the detection of mouse or rabbit
primary antibodies on nitrocellulose or PVDF membranes.
The signal can be visualized using either a chemiluminescent/
chemifluorescent or a chromogenic substrate. Used with the
DuoLuX™ Chemiluminescent/Chemifluorescent Substrate, the
VECTASTAIN® ABC- AmP™ system produces a very high and
sustained light emission signal, with low background, and
permanent fluorescence.
The sensitivity of VECTASTAIN® ABC-AmP™ Reagent combined
with the DuoLux™ substrate allows for the detection of as little
as 1 pg of target protein.
Each VECTASTAIN® ABC-AmP™ kit provides sufficient reagents to develop approximately twenty 100 cm2 blots. Kit consists of
VECTASTAIN® ABC-AmP™ reagent
10x casein solution
biotinylated anti-mouse IgG secondary antibody
DuoLuX™ Chemiluminescent/ Fluorescent Substrate.
Reagents contained in the kits can also be purchased separately.

Western blot visualized by using the VECTASTAIN® ABC-AmP™ kits. Serial dilutions (1:2) of maltose binding protein (MBP) were resolved by electrophoresis on a 12% PAGE reducing gel, transferred onto nitrocellulose membrane and detected with biotinylated anti-MBP made in goat, followed by the VECTASTAIN® ABC-AmP™ Reagent. Blots were developed with [A] DuoLuX™ Substrate using chemiluminescence, or [B] BCIP/NBT substrate.
